Microbiology:

Fresh tissues samples and microbiology samples

Do not mix fresh gastrointestinal organs with visceral organs in the same bag or container. If mixed tissues are submitted in the same container, be sure to list the contents on the label.

Place fresh tissues in whirl pack or Ziploc type bags, double bagged and labeled.

Fresh tissue submissions for microbiological or virology testing should be chilled. Each tissue sample should be packaged separately. Tissue size should be greater than one inch but no greater than five inches in diameter. Swabs of the gastrointestinal tract are useful in testing for aerobic pathogens; however, they are inadequate for testing of the presence of anaerobes. If an anaerobic pathogen is suspected, submit tissues or a segment of the intestine/colon with the ends tied off to ensure an anaerobic environment. The segment should be a minimum of four inches in length. Alternatively, swabs can be sent immersed in anaerobic transport media.

Formalin Fixed Tissue Samples

Tissues submitted for histopathology need to be fixed prior to shipping, preferably in 10% neutral-buffered formalin. Adequate fixative volume is important for proper fixations and should be 10 times greater than the tissue volume. Tissues should not exceed one centimeter in thickness. If formalin is not available, the tissue should be stored refrigerated and shipped cold (gel packs). Do not freeze.

Specific Submission Information

(The procedures listed below are not all inclusive. They represent the tests most commonly requested. For other tests, please call the laboratory at 775-688-1182, ext 231 for details.)

COMMENTS

Acid Fast Stain

Ileum, lymph node, rectal mucosa scraping; tissue impression smear, feces

Sterile container for tissue, slide folder for slides

cold

Same day

Submit 3 slides when submitting tissue impression smears or rectal mucosal scrapings.

Bacterial aerobic culture

Affected tissue, swab, exudate or milk

Sterile container, culturette, with Amies transport media

cold

3-5 days

Identify source site for swabs.  Milk aseptically collected.  A least 2ml of milk required

Bacterial anaerobic culture

Tissue; swab

Anaerobic transport   culturette; sterile container

cold

7-10 days

Identify site for swabs & exudates; animals dead for more than 4-hours are generally unsuitable for any anaerobic cultures due to invading post-mortem invasion of organisms.

Antimicrobial susceptibility

Bacterial isolate

 Bain heart infusion slant

cold

2 days

Infectious substance shipping required

Anthrax

Blood , spleen, eyeball, ocular fluid

Red top vacutainer tube for blood or eyeball fluid, sealed container, spleen or eyeball.

cold

2-5 days (prelim. same day)

Label submitting paper work: Anthrax Suspect; Contact lab before submitting; do NOT send umbilical blood tape

Brucella Culture

Tissue; milk- at least 20ml/ quarter, 100ml composite

Sterile container

cold

7-14 days

Contact lab before submitting. Collect Aseptically.

Campylobacter fetus fetus
(abortions in sheep and occ. cattle)

Stomach contents; fetus; fetal membranes; Placenta

Clark’s or Amies w/charcoal transport  media for washes & swabs. Sterile container for tissues

cold

7 days min.

Samples should reach the lab within 24 hours of collection, in the absence of placenta the likelihood of a diagnosis is severely reduced.

Campylobacter fetus jejuni
(diarrhea)

Freshly passed feces

Sterile container

cold

7 days min.

Samples submitted in gloves/OB sleeves will be refused; Ideally samples should reach the lab within 5 hours of collection

Campylobacter fetus venerealis
(infertility)

Vaginal mucus (from heifer 30-80 days post-breeding); Preputial washing (from bull)

Amies transport media (call ahead to obtain from lab)
Amies transport media (call ahead to obtain from lab)

cool

7 days min.

Washing must be cloudy and blood tinged to show adequate sampling; Do not refrigerate; Transport immediately to lab, must be maintained in microaerophilic atmosphere to survive; Samples should reach the lab within 5 hours of collection

Campylobacter hyointestinalis
(Swine Proliferative Enteritis)

 Affected intestine

. Sterile container

cold

7 day min.

Samples should reach the lab within 5 hours of collection

Campylobacter sputorum mucosalis

 Affected intestine

 Sterile container

cold

7 days min.

Samples should reach the lab within 5 hours of collection

Clostridial FA*

 Deep muscle tissue

 Sterile container

cold

Same day

Specimen must arrive at the lab within 24 hours of collection; Cl. septicum is a frequent post-mortem invader.

Dermatophilus
(Dermatophilosis)

Thick scabs

 Sterile container

cold

3-5 days (prelim. 24hrs)

Use leakproof containers

Fungal Culture

Hair with root (for dermatophytes);. Scab;Tissue (for systemic mycoses);. Fluid (for systemic mycoses)

Sealed paper envelope; Sterile container

cold

14-30 days.

Samples for dermatophyte testing must NOT be submitted in an airtight container; Samples sent with clipped hair are unacceptable; Good collection technique is ESSENTIAL for proper culture results.
COLLECTION TECHNIQUE (for dermatophytes): First CLEAN the area well with alcohol, allow to dry, then collect sample, plucking hair and scraping the edge of lesion; Samples submitted without first disinfecting the area will usually be overgrown by contaminants, resulting in a false negative culture.

Gram Stain

Slide Impression of Tissue; heat fix

Slide folder; Culture tube (or sealed plate)

cold

Same day

For entrotoxemia, estimates of clostridium, intestine smears  should be made within 1-2 hrs postmortem.

Milk Bulk Tank Sample
(for quantitative analysis)

Representative sample

Sterile screw cap or snap top vial

cold

3-5 days

Freeze IMMEDIATELY following collection; screw cap tubes are available for purchase from the lab

Milk Culture

Quarter or combined sample

Sterile screw top or snap top tube

cold

3-5 days

Sterile Collection

Mycobacterium* paratuberculosis
(Johne's)

Fresh feces (at least 3gms, thumb size lump);.intestine (ileocecal junction, ileum, and adjacent lymph nodes are best)

Sterile container or whirlpacks

cold

4 months

Samples submitted in inverted OB sleeves, gloves, or open containers are NOT acceptable; and will be refused; Immediately refrigerate tissue samples. Culture held 15 weeks before considered negative.

Mycoplasma Culture
(bovine only)

Milk; Swab; Fresh tissue

Sterile screw top or snap top tube; Amies with charcoal or culturette. Sterile container for milk

cold

7-21 days

Contact laboratory before submitting to insure media is available.

Urine Culture

Urine collected by cystocentesis or catheter

Sterile leakproof container

cold

3 days

Urine samples collected by cystocentesis or from catheterized animals are preferred, however, mid-stream collections are suitable.
